How to talk about methylation with patients and with professionals — and the final message.
Your DNA is like the instruction manual of your body. Methylation is one of the ways the body decides which instructions should be read more, read less, or kept silent. But methylation is not simply good or bad. If methylation happens in the wrong place, it may silence a protective gene. If methylation is too low in repetitive DNA, the genome may become unstable. The goal is balance: protective genes should remain available, inflammatory genes should not stay chronically active, and repetitive DNA should remain controlled.
DNA methylation must be interpreted according to genomic context, tissue specificity, gene function and clinical correlation. Promoter and enhancer methylation often reduce regulatory accessibility and may lower expression, especially in CpG-rich regulatory elements. Gene body methylation is more complex and may correlate with transcriptional activity, transcript stability and suppression of spurious internal transcription. Repetitive regions such as LINE-1 and Alu elements are often used as indicators of global methylation stability. Interpretation should integrate methylation data with genotype, biomarkers, symptoms, medications, exposures and lifestyle.
